Story: A genetically created Anaconda, cut in half, regenerates itself into two aggressive giant snakes, due to the Blood Orchid.

Picking up immediately where Anaconda 3 left off, herpetologist Amanda Hayes finally learns her employer Murdochs true colors when he hires a doctor to harvest a fresh supply of blood orchids to experiment with the regenerative nectar on a baby snake in order to seek treatment for his own terminal illness. Amanda is forced to secretly bring together another team of scientists whom face off against a heavily armed squad of Murdochs men to steal the coveted orchids before the killer Anaconda hunts both parties down.

In a desperate attempt to cling to life, shortly after the terrifying events of , Murdoch enlists the help of a young doctor to harvest the rejuvenating serum of the rare Blood Orchids. Once more, a failed experiment leads to a new reptilian horror, as an ever-growing, genetically enhanced super-serpent breaks free from the laboratory. Now, the dauntless herpetologist, Amanda Hayes, and a handful of scientists must outsmart Murdochs armed-to-the-teeth henchmen, to have a fighting chance. But, can they put an end to the nightmare of the mutated Anacondas?
